Fluorescence labelling has become increasingly popular as a viable alternative to radiolabelling for producing materials for a variety of binding and enzyme assays, including fluorescence polarisation and FRET techniques. This is partly a result of a desire to move away from using radioactive isotopes but has also been driven by some inherent advantages of fluorescence tags: higher stability, improved spatial resolution, ease of detection.

Whereas radioactive labelling of peptides usually relies on the introduction of a radiolabelled amino acid as part of the natural structure of the peptide, fluorescent tags are introduced as an additional moiety to the molecule. This requires a variety of strategies to be developed to successfully incorporate these labels.

Fluorescent tags are commonly introduced onto amino groups (via a carboxyl or isothiocyanate group of a tag) or onto thiol groups (via a maleimide or iodoacetyl function of the tag). Alternatively, fluorescent labelled amino acids can be incorporated into the peptide during the solid phase synthesis. Whatever your requirements CRB have the capability to prepare your desired peptide.

We can offer peptides labelled with Cyanagen's Chromis dyes. These dyes have excellent stability, are available in cationic, anionic and zwitterion forms with a wide range of reactive functionalities, covering the emission spectrum from 400 to 850 nm. They represent a cost effect alternative to other proprietary dyes as they can be used without royalty payments.

We can also supply peptides labelled with the range of AlexaFluor®, BODIPY® and Oregon green® dyes obtained under licence from Molecular Probes Inc. AlexaFluor® are available at wavelengths covering the spectrum as follows:

We are the exclusive suppliers of CyDye™ labelled peptides which can incorporate any of the range of CyDye™ fluorophores from GE Healthcare (formerly Amersham Bioscience). CyDyes™ are an excellent choice for labelling peptides in a variety of fluorescence based applications including drug screening and assay development.

Cydyes™ are available at wavelengths covering the spectrum as follows:

Examples of other fluorescent groups we can incorporate into peptides include:
  • Fluorescein (FAM or FITC)
  • Rhodamines
  • Edans
  • Amc (7-amino-4-methylcoumarin); Mca (7-methoxycoumarin-4-acetic acid)
  • Dabcyl ; Dabsyl ; Dansyl ; DNP.
