Guidelines for stable-labelled tryptic peptide selection
- 8-20 residues: 800 — 2000 MU
- Contain Leu, Val, Pro, Phe
- Avoid sequences containing Met or Cys. If Cys must to be included, the Cys can be introduced as the Cys(CH2NHCOCH3) derivative, to reflect acetamidomethylation of protein Cys residues with iodoacetamide
- Avoid peptides with C-terminal KK, RR, KR or RK (ragged-ends)
- Avoid peptides containing RP (missed cleavage sites)
- Avoid hydrophobic sequences as they can be difficult to synthesise or have poor solubility
- Avoid peptides containing sites for post-translational modifications
- Check the peptide is unique to your protein (BLAST)
